Why multi-component preparations are formulated rather than combined, what determines whether two compounds are compatible in one solution, and what a blend certificate can establish.

Compatibility is a solution-chemistry question

Two compounds share a solution successfully when both are soluble under the same conditions and neither destabilises the other. Neither can be assumed. Peptides differ substantially in the pH at which they dissolve readily — basic sequences frequently require dilute acid while others are unstable under the same conditions — so a solvent optimal for one component may be poor for the second. Where the optimal conditions for two compounds do not overlap, no formulation satisfies both, and the resulting preparation compromises one or both.

Differential stability in shared conditions

Even where both components dissolve, they may not degrade at comparable rates. Deamidation is strongly pH-dependent and oxidation susceptibility follows residue composition, so a pH selected to keep both compounds in solution may sit well outside the optimum for the stability of one. The consequence is that the ratio between components in a blend is not fixed over time: the less stable component declines faster, and a preparation that was correct at formulation drifts from its stated composition. This is a property of multi-component solutions generally rather than a defect in any particular blend.

Why blends are formulated rather than combined

Combining two separately prepared solutions produces a mixture whose pH, ionic composition, and final concentrations are the incidental result of what was combined, not the result of a decision. A prepared blend is formulated under controlled conditions with the compatibility questions above addressed in advance and the resulting composition analysed. The difference is not that one is more convenient — it is that one has a characterised composition and the other has an assumed one.

Analytical verification of a multi-component preparation

Identity confirmation for a blend addresses each component separately. Mass spectrometry reports the observed masses of the individual compounds present, which is why a blend certificate lists identity results per component rather than a single figure. Chromatographic purity is more complicated: in a multi-component preparation, peaks corresponding to each intended component are all legitimately present, so an area-percent purity figure calculated as though one were the target and the rest impurities does not carry its usual meaning. The certificate should be read for what it reports per component rather than for a single headline number.

What a blend has no single value for

A blend is a mixture rather than a chemical substance, so it has no single CAS number, no single molecular formula, and no single molecular weight. Those fields are correctly absent from a blend record rather than missing from it. Any supplier presenting a single CAS number for a multi-component preparation is presenting the identifier for one component as though it described the whole, which is a category error worth noticing.

What a blend certificate establishes

It establishes what was measured on the analysed lot: which components were detected, their identity against theoretical mass, and the purity of the material as characterised by the issuing laboratory. It does not establish that the ratio between components has been maintained since analysis, which is a stability question addressed by storage conditions and elapsed time rather than by the certificate. As with single-compound material, the certificate describes the lot at the point of analysis, and the record is most informative when read together with the manufacture and analysis dates.
