Temperature Excursion and Assessing Material on Receipt
What transit warmth does to lyophilised material, what can be established by inspection, and what cannot.
What a temperature excursion is
A temperature excursion is a period during which material is held outside its specified storage range. The term describes an event, not an outcome — whether an excursion caused degradation depends on the temperature reached, the duration, and the sequence involved. Brief warming of lyophilised material is a different event from the same warming applied to solution, and treating all excursions as equivalent produces both unnecessary discards and unwarranted confidence.
Why lyophilised material tolerates transit
Most degradation pathways relevant to peptides require water. Oxidation, deamidation, and hydrolysis all proceed substantially faster in solution than in the dry state, which is why lyophilised material is the storage form and why cooling elements in transit packaging are a margin of safety rather than a strict requirement for the dry product. Material that arrives at ambient temperature after transit has not thereby been established as compromised. The relevant question is what the material has experienced cumulatively, not what temperature the package registered on arrival.
What to check on receipt
Confirm that the lot number on the vial matches the lot number on the certificate supplied. Check that the cake is present and adheres to the expected appearance for that compound — a compact cake at the base of the vial rather than material dispersed around the walls, which can indicate the vial was inverted while warm or that the seal was compromised. Confirm the vial seal is intact. Record the arrival condition before storing, because a determination made weeks later cannot reconstruct what arrived.
The limits of visual assessment
Appearance establishes less than is generally assumed. A cake that has shrunk, discoloured, or collapsed indicates a problem, but the absence of those signs does not establish that material is intact. Oxidation and deamidation produce no visible change at levels that matter analytically, and a fully normal-looking vial can carry degradation that only chromatography would detect. Visual inspection is therefore useful for exclusion and not for confirmation: it can identify material that is clearly compromised, and cannot certify material that is not.
Material of uncertain history
Where storage history is unknown or an excursion cannot be characterised, the honest position is that condition is undetermined rather than either acceptable or compromised. For qualitative work the practical risk is often tolerable; for quantitative work it is not, because an unmeasured loss of active material propagates through every subsequent calculation. Re-analysis is the only route to certainty. Material with a documented storage history from receipt onward avoids this problem entirely, which is the argument for recording arrival condition even when nothing appears wrong.

Does a warm package mean the material is degraded?
