What Is a Lyophilized Peptide?
Frozen, then dried under vacuum so the ice turns straight to vapour. Why research peptides are supplied this way, and what the dry powder can and cannot tell you.
Freeze-dried, and why
A lyophilized peptide has been freeze-dried. It is frozen, then dried under vacuum so the ice turns straight to vapour, leaving a dry powder or "cake". Taking out most of the water slows the chemical reactions that break peptides down. That is why research peptides are usually supplied lyophilized and reconstituted only when needed.
The three stages of freeze-drying
Freeze-drying runs in three stages. In freezing, the solution is cooled until its water forms ice. In primary drying, the pressure is lowered and the ice sublimes; this is usually the longest step. In secondary drying, the temperature is raised to remove water that never froze.
Dry is not water-free
The dried material is not completely water-free, and it can take up moisture from the air. Published peptide-handling guidance says moisture greatly reduces long-term stability. It recommends letting a cold vial reach room temperature before opening it, so condensation does not form on the powder.
What the cake can and cannot tell you
A lyophilized cake can look like a solid puck, a loose powder or a thin film, and a small quantity can be hard to see. Appearance does not show identity or purity; those come from HPLC and mass spectrometry on the lot’s certificate. For research use only. Not for human or veterinary use.
